William Shakespearen syntymäpäivästä ei ole säilynyt tarkkaa tietoa, mutta hänet kastettiin synnyinkaupungissaan Stratford-on-Avonissa 26.4.1564 oletettavasti muutaman päivän ikäisenä, ja hänen merkkipäivänään pidetään yleisesti huhtikuun 23:tta. Hän syntyi melko varakkaaseen perheeseen ja kävi paikallista latinakoulua ainakin muutaman vuoden. Vuonna 1582, ollessaan 18-vuotias, hän meni naimisiin 26-vuotiaan Anne Hathawayn kanssa. Perheeseen syntyi kolme lasta, Susanna 1583 ja vuonna 1585 kaksoset, tytär Judith ja myöhemmin 11-vuotiaana kuollut poika Hamnet. Monilta osin Shakespearen vaiheista ja toimista ei ole täsmällistä tietoa, mutta 1590-luvulla hän alkoi saavuttaa mainetta Lontoon teatteripiireissä. Ensimmäiset painetut teokset olivat runoelmia, ja useimmat näytelmät ilmestyivät painettuina vasta tekijän kuoleman jälkeen kootuissa teoksissa, ns. ensimmäisessä foliopainoksessa 1623. Teattereissa niitä kuitenkin esitettiin paljon, ja uusia näytelmiä syntyi tiuhaan, joskus kolmekin vuodessa. Ensimmäinen Shakespearen kirjoittamaksi tiedetty näytelmä on vuonna1592 esitetty Henrik VI. Shakespeare kuului myös teatteriseurueisiin ja oli yksi Globe-teatterin omistajista. Todennäköisesti viimeinen näytelmä on 1611 ensiesityksensä saanut Myrsky, mutta senkin jälkeen hän kirjoitti muutamia näytelmiä yhteistyössä toisten näytelmäkirjailijoiden kanssa. 1610-luvulla hän jätti Lontoon ja teatterin ja vetäytyi Stratfordiin, missä hän kuoli 52-vuotiaana 23.4.1616.
